Ülevaade
Metalloproteoomika uurimisgrupp keskendub järgmistele teadusteemadele: (i) bioloogiliselt oluliste metalliioonide tsingi, vase ja raua homeostaasis osalevate valkude struktuursed ja funktsionaalsed uuringud kasutades erinevaid tehnoloogiaid nagu NMR, ESI MS, ICP MS, fluorestsents ja UV-VIS spektroskoopia, (ii) biometallide tsingi, vase ja raua roll amüloidsete peptiidide fibrillisatsioonile ja in vitro ning in vivo toksilisusele, mis on seotud Alzheimeri tõvega, (iii) uute metallioone kelateerivate ligandide väljatöötamine ja nende rakendamine Wilsoni ja Alzheimeri tõve ravimstrateegiate väljatöötamiseks.
Tähtsamad tulemused
Metalloproteoomika uurimisrühma viimaste aastate väljapaistvamad teadustulemused:Kasutades rühma poolt väljatöötatud LC-ICP MS metoodikat püüti määrata Cu(II)-sidumise afiinsused Cu,Zn-SOD1 ja selle ALS-i haigust põhjustava mutandi jaoks. Selgus, et seostumise termodünaamiliste konstantide määramine on raskendatud metalliioonide kõrge kineetilise inertsuse tõttu.Kasutades fluorestsentstiitrimist määrati Hg(II) ioonide sidumisafiinsused ApoE2, E3 ja E4 valkude suhtes.Näidati, et mikromolaarne lipoehappe (LA) lisamine taastab kahjustatud raku morfoloogia Wilsoni tõbe modelleerivates 3T3-L1 ATP7A-/- rakkudes. Täiendavad LA mõju uuringud selgitasid LA mõju raku redokspotentsiaalile ja selenoproteiinide ekspressiooni ülesregulatsioonile. Samuti uuriti võrdlevalt teisi vase kelaatoreid nagu BCS.
